Genetic diversity and population structure of Brazilian native bovine breeds PAB
Serrano,Germana Maria; Egito,Andréa Alves do; McManus,Concepta; Mariante,Arthur da Silva.
Conservation and improvement strategies should be based on the association between genetic and phenotypic characteristics. The objective of this work was to characterize five native Brazilian cattle breeds (Caracu, Crioulo Lageano, Curraleiro, National Polled and Pantaneiro) and two commercial breeds (Holstein and Nellore) using RAPD technique to estimate genetic distances and variability between and within breeds. Genetic relationships were investigated using 22 primers which generated 122 polymorphic bands. Analysis of molecular variance indicated that most of the genetic variation lay among individuals within populations. The genetic variabilities between pairs of breeds were statistically significant. The smallest genetic divergence was between Crioulo...

Genetic diversity of the Brazilian Creole cattle Pé-duro assessed by microsatellites and mitochondrial DNA R. Bras. Zootec.
Oliveira,Ana Paula Ferreira de; Carvalho,José Herculano de; Miretti,Marcos Mateo; Lara,Maria Aparecida Cassiano; Contel,Eucleia Primo Betioli.
The objective of this study was to describe the genetic diversity and structure of the largest Pé-duro population by assessing variation at ten autosomal microsatellite (STR) loci and mitochondrial DNA (mtDNA) sequences. The mean expected heterozygosity was 0.755, the mean observed heterozygosity was 0.600 and significant inbreeding coefficient (Fis) and deviations from the Hardy-Weinberg equilibrium in most of analyzed loci demonstrate the impact of inbreeding and homozygosis on this population. A more in-depth genetic analysis could be achieved by expanding the STR list. The analysis of mtDNA provided evidence of ancestral African taurine haplotypes in Pé-duro and excluded maternal Zebuine introgression. In this report, the main Pé-duro population is...

Genotype and subtype analyses of Cryptosporidium isolates from cattle in Hungary OAK
Plutzer, Judit; Karanis, Panagiotis.
Seventy-nine faecal samples from calves with diarrhoea were collected on 52 farms from different counties in Hungary to investigate the species, genotypes and subtypes of Cryptosporidium in calves. Oocysts from faecal sample collected from each animal were concentrated using sucrose gradient centrifugation and examined by immunofluorescence assay (IFA). Genomic DNA was extracted from microscopically positive samples and nested PCR was performed to amplify the partial SSU rRNA and GP60 genes of Cryptosporidium that were subsequently digested by SspI, VspI and MboII restriction enzymes to determine the Cryptosporidium species and genotype present. Cryptosporidium parvum was detected in 21 samples while the Cryptosporidium deer-like genotype was found in...

Theileria orientalis is a benign protozoan species that is widely distributed in Japan, yet sometimes causes serious economic losses in the livestock industry. In this study, we conducted a molecular survey based on genes encoding the major piroplasm surface protein (MPSP) and p23 for T. orientalis detected in cattle grazing in southern areas of Japan, consisting of 2 farms in Kumamoto prefecture (Aso and Kuma districts) and 3 farms in Okinawa prefecture (Ishigaki, Iriomote, and Yonaguni Islands). High prevalence rates of T. orientalis infection were shown in all the cattle populations using the diagnostic MPSP- and p23-PCR assays. Phylogenetic analyses revealed 4 MPSP genotypes and 3 p23 genotypes. Furthermore, MPSP genotype-specific PCR methods were...

Genotyping of Clostridium perfringens associated with sudden death in cattle J. Venom. Anim. Toxins incl. Trop. Dis.
Miyashiro,S; Baldassi,L; Nassar,AFC.
Toxigenic types of Clostridium perfringens are significant causative agents of enteric disease in domestic animals, although type E is presumably rare, appearing as an uncommon cause of enterotoxemia of lambs, calves and rabbits. We report herein the typing of 23 C. perfringens strains, by the polymerase chain reaction (PCR) technique, isolated from small intestine samples of bovines that have died suddenly, after manifesting or not enteric or neurological disorders. Two strains (8.7%) were identified as type E, two (8.7%) as type D and the remainder as type A (82.6%). Commercial toxoids available in Brazil have no label claims for efficacy against type E-associated enteritis; however, the present study shows the occurrence of this infection. Furthermore,...

Granulomatous myelitis associated with hemorrhagic syndrome due to consumption of Vicia villosa by cattle Ciência Rural
Sonne,Luciana; Raymundo,Djeison Lutier; Antoniassi,Nadia Aline Bobbi; Bandarra,Paulo Mota; Almeida,Paula Rodrigues de; Vaz Júnior,Itabajara da Silva; Driemeier,David.
An unusual case of spontaneous Vicia villosa poisoning affected a 6-year-old Holstein cow. Although the most striking findings included a generalized hemorrhagic condition associated with granulomatous myelitis, histological lesions typically seen with the vetch-associated systemic granulomatous syndrome were also present. Prominent gross findings were bloody nasal and oral discharges, disseminated hemorrhages, and bloody feces. Generalized hemorrhages associated with infiltration of numerous organs by lymphocytes, plasma cells, macrophages, multinucleated giants cells, and eosinophils were the main microscopic findings. Anti-CD68 immunostaining confirmed the presence of moderate histiocytic infiltrate and multinucleated giant cells in the bone marrow....

Heat identification by 17β-estradiol and progesterone quantification in individual raw milk samples by enzyme immunoassay Electron. J. Biotechnol.
Domènech,Anna; Pich,Sara; Arís,Anna; Plasencia,Carmen; Bach,Alex; Serrano,Alicia.
Background: There is a substantial decline in first-service-pregnancy-rate in dairy cows. In this regard, future prospects are to measure milk hormones on-farm and progesterone levels in milk are not enough to precise ovulation unless connected to other data. The objectives of this study were to investigate whether 17β-estradiol could be measured from individual cow milk samples using a commercially available non-radiolabelled enzyme immunoassay kit (EIA) with no previously reported milk application, and whether those detections could precisely illustrate 17β-estradiol pre-ovulation peak in spite of its limited concentration and short manifestation in milk. Results: Milk sample treatments for progesterone and 17β-estradiol EIA...

Ybañez, Adrian Patalinghug; Ybañez, Rochelle Haidee D.; Claveria, Florencia G.; Cruz-Flores, Mary Jane; Xuenan, Xuen; Yokoyama, Naoaki; Inokuma, Hisashi.
A total of 658 cattle in 6 provinces in the Philippines were screened for Anaplasma marginale infection by using a diagnostic heat-shock operon (groEL) gene-PCR assay. The screening-positive samples were further tested using the major surface antigen protein 1a (Msp1a) gene-PCR assay. Screening PCR results showed 130 cattle (19.8%) were positive for the A. marginale infection. Subsequent amplification using the Msp1a gene only showed 93 samples (14.1%) to be positive. In addition, 37 tandem-repeat structures, including 20 novel structures, and 41 distinct genotypes were identified. Interestingly, multiple infections of 4 different genotypes were also observed in A. marginale-infected cattle. The present study demonstrated the prevalence and...

HPLC identification of isoniazid residues in bovine milk Arq. Bras. Med. Vet. Zootec.
Leite,R.M.H.; Leite,R.C.; Lima,J.A.; Fóscolo,C.B.; Mota,P.M.P.C.; Lobato,F.C.F.; Lage,A.P..
The high pressure liquid chromatography (HPLC) was used for the identification of isoniazid (isonicotinic acid hydrazide) in the milk of cattle treated with a dose of 25 mg/kg/day in alternated days. The effect of milk pasteurization on the isoniazid residue concentration was also studied. The drug excretion presented a cyclic variation, with higher levels in the first day after administration (aa), a mean of 1104.48µg/l, and a decrease two days aa, with a mean of 104.12µg/l. Four days after the last administration of the drug it was not possible to identify residues of isoniazid in the milk of treated animals. Body weight and milk yield influenced the amount of the excreted drug, and pasteurization decreased (mean 47.07%) the concentration of isoniazid...

Impact of Beef Quality on Market Signals Transmitted by Grid Pricing AgEcon
Johnson, Heather C.; Ward, Clement E..
Value-based marketing is shaping cattle production; however, market signals differ dramatically depending on carcass quality. This study applies a two-stage coefficients of separate determination procedure to four regional fed cattle datasets sorted by grid value and by carcass quality attributes. Weight is the strongest signal sent when higher valued cattle and better quality cattle are sold on a grid. Quality characteristics send stronger signals when lower valued cattle and poorer quality cattle are sold on a grid. Producers of lower quality cattle can potentially gain $52 to $149 per head by improving quality and $113 to $150 per head by adding weight.

Implementation of a parentage control system in Portuguese beef-cattle with a panel of microsatellite markers Genet. Mol. Biol.
Carolino,Inês; Sousa,Conceição O.; Ferreira,Sónia; Carolino,Nuno; Silva,Fátima S.; Gama,Luís T..
A study was conducted to assess the feasibility of applying a panel of 10 microsatellite markers in parentage control of beef cattle in Portugal. In the first stage, DNA samples were collected from 475 randomly selected animals of the Charolais, Limousin and Preta breeds. Across breeds and genetic markers, means for average number of alleles, effective number of alleles, expected heterozygosity and polymorphic information content, were 8.20, 4.43, 0.733 and 0.70, respectively. Enlightenment from the various markers differed among breeds, but the set of 10 markers resulted in a combined probability above 0.9995 in the ability to exclude a random putative parent. The marker-set thus developed was later used for parentage control in a group of 140 calves from...
